Sample Submission Requirements
Sample Preparation for the AGRF Microarray Labelling and Hybridisation Services
1 All total RNA samples submitted are required to have the following:
a. Any RNA extraction process is sufficient, however it is a requirement that the total RNA is passed through a column purification step after extraction.
b. The total RNA is only resuspended in water.
c. For RNA samples that are shipped as a lyophilised sample, these will be re-hydrated before processing by AGRF. For samples originating outside of Australia, lyopholisation of samples is recommended prior to shipping to AGRF.
d. Each total RNA sample must be run on a gel or Bioanalyser, in order to ensure that the sample was in optimal condition prior to being sent to the AGRF. It is advisable to include an image of the total RNA in the package to be sent to the AGRF.
e. On arrival at the AGRF, every sample will undergo a QC check to ensure that the sample will label to maximum efficiency.
f. In the event that a sample fails the AGRF QC check, AGRF will notify the customer. It will then be at the discretion of the customer as to whether the labelling of the sample will proceed.
2 Requested total RNA weights for the relevant platforms are:
|
Platform |
Total RNA Weight |
|
Affymetrix GeneChip |
15μg (7μg Labelled) |
|
Affymetrix All Exon |
7μg (3μg for rRNA reduction, of which 1μg for labelling |
|
Affymetrix Gene 1.0ST |
1μg (100ng for labelling) |
|
illumina Sentrix |
1 - 3μg (500ng for labelling) |
|
Combimatrix 4 x 2k |
5μg (2.5μg for labelling) |
|
Combimatrix 12k |
10μg (2.5μg for labelling) |
3 A necessary requirement for sending total RNA samples to Australia is that an appropriate Australian Quarantine Inspection Service document is included in the shipment, as this will expedite the sample arrival process. (see attached pdf).
